Biological Background: TRAF6BP/TAX1BP1 Function and Localization
- Tax1-binding protein 1 (TAX1BP1, also known as TRAF6-binding protein, T6BP, TXBP151, CALCOCO3) is a ubiquitin-binding adapter protein that participates in inflammatory, antiviral, innate immune processes, and selective autophagy.
- Functions as a key negative regulator of NF-kB and IRF3 signaling pathways by acting as an adapter for the ubiquitin-editing enzyme A20/TNFAIP3 to bind and inactivate its substrates, such as TRAF6 and RIPK1, thereby disrupting IL-1 and TNF-induced NF-kB signaling. Related references: PMID:17703191
- Disrupts the interactions between the E3 ubiquitin ligase TRAF3 and TBK1/IKBKE to attenuate 'Lys63'-linked polyubiquitination of TBK1 and subsequent IFN-beta production. Related references: PMID:21885437
- Acts as a macroautophagy/autophagy receptor facilitating the xenophagic clearance of pathogenic bacteria like Salmonella typhimurium and Mycobacterium tuberculosis, and recruits RB1CC1 to SQSTM1-ubiquitin condensates to promote their autophagic degradation. Related references: PMID:26451915 PMID:33226137 PMID:34471133
- Mediates the autophagic degradation of other substrates, including TICAM1, and inhibits virus-induced apoptosis by inducing 'Lys-48'-linked polyubiquitination and degradation of MAVS via recruitment of the E3 ligase ITCH. Related references: PMID:28898289 PMID:27736772
- Subcellular localization includes cytoplasm, mitochondria, preautophagosomal structures, cytoplasmic vesicles, and autophagosomes.
- Ubiquitously expressed in all tissues tested.
- Contains a zinc-finger domain and coiled-coil regions; subject to phosphorylation and alternative splicing.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the C-terminal region (aa 709-789), which may influence epitope accessibility; consider validating the antibody in the relevant sample system.
- For IHC-P, optimize antigen retrieval conditions and validate with known positive and negative human tissue controls. Rat cross-reactivity offers potential for translational models.
- For ELISA, the polyclonal nature may provide sensitive detection; pairing with a monoclonal antibody could be evaluated for sandwich assay development.
- Verify the observed molecular weight (~91kDa) in your experimental system, as post-translational modifications or isoforms may shift migration.
