Biological Background: TRAF6BP/TAX1BP1 Function and Localization

  • Tax1-binding protein 1 (TAX1BP1), also known as TRAF6-binding protein or T6BP, is a ubiquitin-binding adapter protein encoded by the TAX1BP1 gene. It participates in inflammatory, antiviral, and innate immune processes, as well as selective autophagy regulation (PubMed:29940186, PubMed:30459273, PubMed:30909570). Related references: PMID:29940186, PMID:30459273, PMID:30909570

  • TAX1BP1 negatively regulates NF-kappa-B and IRF3 signaling by acting as an adapter for the ubiquitin-editing enzyme A20/TNFAIP3, leading to deubiquitination of substrates like TRAF6 and RIPK1, thus inhibiting IL-1 and TNF-induced NF-kB pathways (PubMed:17703191). Related references: PMID:17703191

  • It disrupts the interaction between TRAF3 and TBK1/IKBKE, attenuating 'Lys63'-linked polyubiquitination of TBK1 and thereby reducing IFN-beta production (PubMed:21885437). Related references: PMID:21885437

  • As an autophagy receptor, TAX1BP1 facilitates xenophagic clearance of pathogenic bacteria such as Salmonella typhimurium and Mycobacterium tuberculosis (PubMed:26451915). Upon NBR1 recruitment to SQSTM1-ubiquitin condensates, it acts as the major recruiter of RB1CC1 to promote autophagic degradation (PubMed:33226137, PubMed:34471133). Related references: PMID:26451915, PMID:33226137, PMID:34471133

  • TAX1BP1 also mediates the autophagic degradation of other substrates including TICAM1 (PubMed:28898289). Related references: PMID:28898289

  • Subcellular localization includes cytoplasm, mitochondria, preautophagosomal structures, and autophagosomes.

  • It is widely expressed across all tested tissues.

  • The protein undergoes alternative splicing and phosphorylation; it contains zinc-finger domains and coiled-coil regions.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to a synthetic peptide from amino acids 690–789 of human TRAF6BP/TAX1BP1. Verify epitope conservation in your target species (cross-reactivity confirmed for human and mouse).
  • For Western blotting, expect a band near 91 kDa. Use appropriate positive controls (e.g., cell lines expressing TAX1BP1) and loading controls.
  • For ELISA, optimize coating conditions and antibody titers. Validate with known standards or recombinant protein.
  • Multiple isoforms exist due to alternative splicing; confirm that the antibody detects the desired isoform in your system.
  • The antibody has not been validated for immunoprecipitation or immunofluorescence; if needed, test under your specific conditions.
