Biological Background: TRAF6 Function and Localization
- TRAF6 (TNF receptor-associated factor 6) is a member of the TRAF family of adapter proteins and E3 ubiquitin ligases.
- It catalyzes the synthesis of Lys-63-linked polyubiquitin chains on target proteins including ECSIT, IKBKG, IRAK1, AKT1, and AKT2.
Related references: PMID:11057907, PMID:18347055, PMID:19465916 - This ubiquitination activity leads to the activation of the NF-kB and JUN signaling pathways.
Related references: PMID:16378096, PMID:17135271, PMID:17703191 - TRAF6 represses c-Myb-mediated transactivation in B-lymphocytes.
Related references: PMID:18093978, PMID:18758450 - It functions as an adapter in signal transduction initiated via TNF receptor, IL-1 receptor, and IL-17 receptor.
Related references: PMID:12140561, PMID:19825828, PMID:8837778 - The protein localizes to the cytoplasm, cell cortex, nucleus, and lipid droplets, and is broadly expressed in tissues including heart, brain, and skeletal muscle.
- TRAF6 contains a RING finger domain, multiple zinc fingers, and a coiled-coil region, and is implicated in osteogenesis and immune regulation.
Experimental Guidance and Technical Tips
- For Western blot (WB), the expected band is approximately 60 kDa; consider loading appropriate positive controls and validating in physiologically relevant cell or tissue lysates.
- In immunofluorescence/immunocytochemistry (IF/ICC), optimize fixation and permeabilization protocols to detect TRAF6 in its multiple subcellular locations (cytoplasm, nucleus, lipid droplets).
- The antibody is reported to cross-react with mouse and rat TRAF6; confirm sequence homology and validate species-specific reactivity in pilot experiments before large-scale use.
- As the immunogen is a synthetic peptide, reactivity may be limited to the immunizing region; consider testing in the context of native and denatured protein conformations across intended applications.
