Biological Background: TRAF2 Function and Localization
- TRAF2 (TNF receptor-associated factor 2), also known as TRAP3, is an E3 ubiquitin-protein ligase that plays a central role in the regulation of cell survival and apoptosis by mediating activation of NF-kappa-B and JNK. Related references: PMID:10346818, PMID:11784851, PMID:12917689
- It catalyzes 'Lys-63'-linked ubiquitination of target proteins such as BIRC3, IKBKE, MLST8, RIPK1, and TICAM1, influencing diverse signaling cascades. Related references: PMID:23453969, PMID:28489822
- As an essential constituent of several E3 ubiquitin-protein ligase complexes, TRAF2 facilitates the ubiquitination of target proteins by bringing them into contact with other E3 ligases. Related references: PMID:15383523, PMID:18981220
- TRAF2 regulates BIRC2 and BIRC3 protein levels by inhibiting their autoubiquitination and subsequent degradation, a function independent of its RING-type zinc finger domain. Related references: PMID:11907583, PMID:19506082
- It promotes 'Lys-63'-linked ubiquitination of MLST8 to regulate mTORC1 and mTORC2 assembly, inhibiting mTORC2 formation while facilitating mTORC1 assembly. Related references: PMID:28489822
- TRAF2 mediates activation of NF-kappa-B by EIF2AK2/PKR, and is required for normal antibody isotype switching from IgM to IgG. Related references: PMID:15121867
- Localized to the cytoplasm, TRAF2 contains characteristic domains including a RING-type zinc finger and a coiled coil region, and is subject to phosphorylation and acetylation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1-300) of human TRAF2, which encompasses the RING domain. This region is highly conserved, consistent with the antibody's cross-reactivity with mouse and rat.
- For Western blotting, a band around 56 kDa is expected. Multiple bands may appear due to post-translational modifications or alternative splicing isoforms; consider validating with appropriate controls.
- For immunohistochemistry (IHC-P), standard antigen retrieval procedures are recommended. Optimize antibody dilution and incubation conditions using positive and negative control tissues.
- For ELISA, titrate the antibody to determine the optimal working concentration for your specific assay format and sample type.
- Since TRAF2 is involved in multiple signaling complexes, co-immunoprecipitation or proximity ligation assays could be explored to study interaction partners, but these applications have not been validated with this product and require independent optimization.
