Biological Background: TPPP/p25 Function and Localization
- TPPP/p25 (p24, p25-alpha, TPPP1) is a 219-amino-acid protein encoded by the TPPP gene, with a molecular weight of approximately 24 kDa.
- Acts as a microtubule nucleation factor in oligodendrocytes, localizing to the Golgi outpost to promote elongation of the myelin sheath; required for uniform polarized growth of distal microtubules and directing proximal process branching. Related references: PMID:31522887 PMID:33831707
- Exhibits microtubule bundling and stabilization activity, maintaining microtubule network integrity. Related references: PMID:17105200 PMID:17693641 PMID:18028908
- Promotes tubulin acetylation by inhibiting the deacetylase HDAC6, thereby regulating microtubule dynamics and stability. Related references: PMID:20308065 PMID:23093407
- Regulates cell migration: phosphorylation by ROCK1 impairs HDAC6 interaction, reducing tubulin acetylation and increasing cell motility. Related references: PMID:23093407
- Involved in cell cycle progression by regulating the G1/S-phase transition. Related references: PMID:23355470
- Participates in mitotic spindle organization and orientation during early mitosis, regulated by LIMK2 phosphorylation. Related references: PMID:22328514
- Possesses magnesium-dependent GTPase activity; functional significance remains under investigation. Related references: PMID:21316364 PMID:21995432
- Subcellularly localizes to the Golgi outpost, cytoplasm, cytoskeleton (microtubule organizing center, spindle), and nucleus; widely expressed across tissues.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide corresponding to amino acids 100–200 of human TPPP/p25 (O94811). For Western blotting, the predicted molecular weight is 24 kDa; consider optimizing gel percentage and transfer conditions accordingly.
- For IHC-P, validate staining in tissue sections with known TPPP/p25 expression and include appropriate controls.
- The antibody cross-reacts with mouse and rat species; verify reactivity and specificity in the relevant sample system for your experimental design.
- ELISA applications may require titration of coating antigen and primary antibody concentration to achieve optimal signal-to-noise ratios.
