- Tropomyosin alpha-3 chain (TPM3), also known as gamma-tropomyosin, tropomyosin-3, and tropomyosin-5 (hTM5), belongs to the tropomyosin family of actin-binding proteins.
- It binds to actin filaments in both muscle and non-muscle cells, playing a central role in calcium-dependent regulation of striated muscle contraction via the troponin complex.
- In smooth muscle, TPM3 regulates contraction through interaction with caldesmon.
- In non-muscle cells, TPM3 is implicated in stabilizing cytoskeleton actin filaments.
- The protein localizes to the cytoplasm and cytoskeleton.
- Alternative splicing generates multiple isoforms, and the protein undergoes post-translational modifications including acetylation and phosphorylation.
- Mutations in TPM3 are associated with nemaline myopathy, and chromosomal rearrangements involving this gene have been linked to proto-oncogenic activity.
