Biological Background: TPM3 Function and Localization

  • Tropomyosin alpha-3 chain (TPM3), also known as gamma-tropomyosin, tropomyosin-3, and tropomyosin-5 (hTM5), belongs to the tropomyosin family of actin-binding proteins.
  • It binds to actin filaments in both muscle and non-muscle cells, playing a central role in calcium-dependent regulation of striated muscle contraction via the troponin complex.
  • In smooth muscle, TPM3 regulates contraction through interaction with caldesmon.
  • In non-muscle cells, TPM3 is implicated in stabilizing cytoskeleton actin filaments.
  • The protein localizes to the cytoplasm and cytoskeleton.
  • Alternative splicing generates multiple isoforms, and the protein undergoes post-translational modifications including acetylation and phosphorylation.
  • Mutations in TPM3 are associated with nemaline myopathy, and chromosomal rearrangements involving this gene have been linked to proto-oncogenic activity.

Experimental Guidance and Technical Tips

  • Based on the immunogen design (recombinant protein corresponding to amino acids 1-284 of human TPM3), this polyclonal antibody is expected to detect both full-length and truncated TPM3 isoforms.
  • In Western blot, a band at approximately 33 kDa is anticipated; consider using whole-cell lysates from human or mouse tissues/cells.
  • The antibody cross-reacts with mouse TPM3, making it suitable for studies in mouse models; however, validation in the specific experimental system is recommended.
  • For ELISA, the antibody can be used as a capture or detection reagent; appropriate pairing and optimization with other TPM3 antibodies or antigens should be performed.
  • Consider testing the antibody in additional applications such as immunoprecipitation or immunofluorescence with appropriate validation.
