Biological Background: TP73 Function and Localization
- Tumor protein p73, also known as p53-like transcription factor or p53-related protein, is encoded by the TP73 gene and belongs to the p53 family.
- Functions in the apoptotic response to DNA damage; isoforms containing the transactivation domain promote apoptosis, while those lacking it act anti-apoptotically and inhibit p53 and transactivating p73 isoforms.
- Acts as a tumor suppressor and an activator of FOXJ1 expression, essential for the positive regulation of lung ciliated cell differentiation. Related references: PMID:34077761
- Localizes to both the nucleus and cytoplasm.
- Expressed in various tissues including brain, kidney, placenta, colon, heart, liver, spleen, skeletal muscle, prostate, thymus, and pancreas; highly expressed in fetal tissue and respiratory epithelium. Related references: PMID:34077761
- Keywords indicate involvement in transcription regulation, cell cycle, apoptosis, and roles as a tumor suppressor; subject to alternative splicing and promoter usage, with metal-binding and zinc finger domains.
- Phosphoprotein with potential isopeptide bond and Ubl conjugation, reflecting complex post-translational regulation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the C-terminal region (aa 487-636) of human TP73, which may recognize both transactivation-competent and -deficient isoforms, but validation in the relevant isoform context is advised.
- For Western blotting, the predicted molecular weight is approximately 70 kDa; use a protein ladder and positive control lysates (e.g., HeLa, MCF7) to confirm the band.
- For immunofluorescence and immunohistochemistry, anticipate nuclear and cytoplasmic staining patterns; permeabilization and blocking conditions should be optimized for each sample type.
- In ELISA applications, titration of both antibody and antigen is recommended to establish optimal signal-to-noise ratios; the antibody can be explored for both direct and sandwich ELISA formats.
- Cross-reactivity with mouse is indicated; for mouse tissue or cell lines, validate under your specific experimental conditions to ensure specificity.
