Biological Background: TP53BP2 Function and Localization
- Encoded by the TP53BP2 gene, apoptosis-stimulating of p53 protein 2 (ASPP2) is a 126 kDa protein also referred to as Bbp, 53BP2, or p53BP2.
- Enhances the DNA binding and transactivation activity of TP53 on proapoptotic gene promoters, thereby promoting apoptosis. Related references: PMID:12524540
- Inhibits NAE1-mediated conjugation of NEDD8 to CUL1, decreasing NAE1-driven apoptosis.
- Regulates cell cycle progression by impeding the G2/M transition.
- Localizes to the cytoplasm, perinuclear region, and nucleus.
- Widely expressed in tissues, with high levels in spleen, thymus, prostate, testis, ovary, small intestine, colon, and peripheral blood leukocytes; expression is reduced in breast carcinomas harboring wild-type TP53 and elevated in lung cancer cell lines.
- Contains structural motifs including ANK repeats and an SH3 domain, and is regulated by phosphorylation.

- For IHC-P, consider optimizing antigen retrieval protocols and validating in formalin-fixed, paraffin-embedded tissue sections known to express TP53BP2, such as spleen or lung tumor samples.
- When performing ELISA, ensure the coating antigen contains the relevant epitope region, and include appropriate negative and positive controls to confirm specificity.
