Biological Background: Topo IIα Function and Localization
- DNA topoisomerase 2-alpha (Topo IIα), encoded by the TOP2A gene, is a critical ATP-dependent enzyme that modulates DNA topology by transiently cleaving and religating double-stranded DNA, allowing the passage of one duplex through another (PubMed:17567603, PubMed:18790802, PubMed:22013166). Related references: PMID:17567603 PMID:18790802 PMID:22013166
- Topo IIα is predominantly localized to the nucleus, with enrichment in the nucleoplasm and nucleolus; it is also detected in the cytoplasm, particularly during cell division.
- It is widely expressed in proliferative tissues such as tonsil, spleen, lymph node, thymus, and testis, and is notably overexpressed in high-grade lymphomas, squamous cell lung tumors, and seminomas (PubMed:9155056). Related references: PMID:9155056
- Post-translational modifications, including phosphorylation, acetylation, and ubiquitination, regulate Topo IIα activity, stability, and cellular localization.
- The protein exhibits alternative splicing, yielding multiple isoforms with molecular weights in the range of 174 to 182 kDa.
- Topo IIα may also participate in circadian rhythm regulation by influencing the period length of BMAL1 transcriptional oscillation (By similarity).
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from the C-terminal region (aa 1431-1531), which may provide high specificity for Topo IIα.
- For Western blot, due to the large protein size (~174-182 kDa), optimize protein transfer conditions (e.g., lower methanol percentage, extended transfer time) and verify bands with appropriate molecular weight standards.
- In immunohistochemistry (IHC-P), consider antigen retrieval protocols (e.g., heat-induced epitope retrieval) and titrate the antibody to minimize background.
- For ELISA applications, this polyclonal antibody can be used as a capture or detection reagent; pairing with a monoclonal antibody may enhance specificity in sandwich assays.
- Cross-reactivity is validated for human samples; for other species, sequence homology of the immunogen should be checked before use.
