Biological Background: TOM70 Function and Localization
- TOM70 (Mitochondrial import receptor subunit TOM70), also known as TOMM70A or translocase of outer membrane 70 kDa subunit, is encoded by the TOMM70 gene (alternative names: KIAA0719, TOM70).
- Acts as a receptor of the preprotein translocase complex of the outer mitochondrial membrane (TOM complex), mediating the import of mitochondrial precursor proteins.

PMID:25609812 - Localizes to the mitochondrial outer membrane, where it functions as a key component of the protein import machinery.
- Subject to various post-translational modifications including acetylation, phosphorylation, methylation, and ubiquitin-like conjugation, which may regulate its activity and interactions.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 245–608 of human TOM70, a region that excludes the N-terminal transmembrane domain; the antibody is expected to recognize the cytoplasmic domain.
- For Western blotting, the predicted molecular weight of TOM70 is ~67 kDa (UniProt: 67.5 kDa), but post-translational modifications may result in a band shift; validate using appropriate controls.
- For immunofluorescence/ICC, verify mitochondrial localization by co-staining with a mitochondrial marker to confirm specificity.
- For ELISA, consider using a recombinant TOM70 protein or peptide as coating antigen; optimize the dilution in your assay system.
- Since this is a polyclonal antibody, lot-to-lot consistency should be verified and the optimal working dilution determined empirically in the relevant sample type.
