Biological Background: TOM20 Function and Localization
- TOMM20 (also known as TOM20, MAS20, MOM19) is the human homolog of the mitochondrial import receptor subunit TOM20, a core component of the translocase of the outer membrane (TOM) complex essential for mitochondrial protein import. Related references: PMID:40080546
- Together with TOMM22, it acts as the transit peptide receptor at the mitochondrial surface, recognizing and guiding cytosolically synthesized preproteins into the TOM40 translocation pore. Related references: PMID:18331822
- TOMM20 associates with VDAC2 and PINK1 kinase at depolarized mitochondria, stabilizing PINK1 to trigger mitophagy via recruitment of the E3 ubiquitin ligase PRKN. Related references: PMID:40080546
- It is required for the translocation of cytochrome P450 monooxygenases across the mitochondrial outer membrane.
- TOMM20 localizes to the mitochondrial outer membrane and contains a single transmembrane helix.
- Post-translational modifications include phosphorylation, isopeptide bond formation, and ubiquitin conjugation (Ubl).
- The protein has a molecular weight of approximately 16.3 kDa and is classified under reference proteome.
Experimental Guidance and Technical Tips
- Since the antibody is raised against recombinant human TOMM20, it may recognize the native protein in WB, IF, and IHC applications. For western blotting, expect a band near 16 kDa; consider using mitochondrial-enriched lysates to enhance signal.
- For immunofluorescence, anticipate mitochondrial outer membrane staining; co-staining with mitochondrial markers is suggested.
- When using for immunohistochemistry, optimize antigen retrieval conditions and validate with appropriate tissue controls.
- The antibody reacts with human, mouse, and rat samples; however, cross-reactivity in other species should be verified experimentally.
- For ELISA, use the antibody as capture or detection reagent and optimize coating conditions.
