Biological Background: TOM20 Function and Localization

  • Also known as Mitochondrial import receptor subunit TOM20 homolog, Mitochondrial 20 kDa outer membrane protein, and Outer mitochondrial membrane receptor Tom20. Encoded by the TOMM20 gene (KIAA0016) in humans.
  • Serves as the central receptor of the translocase of the outer membrane (TOM) complex, essential for recognition and translocation of cytosolically synthesized mitochondrial preproteins (PubMed:40080546). Together with TOMM22, it facilitates preprotein movement into the TOM40 translocation pore (PubMed:18331822). At depolarized mitochondria, it associates with VDAC2 and PINK1, stabilizing PINK1 and triggering PRKN-mediated mitophagy (PubMed:40080546). Related references: PMID:40080546 PMID:18331822
  • Required for the translocation of cytochrome P450 monooxygenases across the mitochondrial outer membrane (By similarity).
  • Localized exclusively to the mitochondrion outer membrane.
  • Post-translationally modified by phosphorylation and isopeptide bond formation (Ubl conjugation).
  • Belongs to the Tom20 family, a group of single-pass transmembrane proteins critical for mitochondrial protein import.

Experimental Guidance and Technical Tips

  • The immunogen (aa 20-145) covers most of the cytoplasmic domain of human TOM20, likely providing good specificity for native protein detection in applications like Western blot and immunofluorescence.
  • For Western blot, the theoretical molecular weight is ~16 kDa (UniProt: 16298 Da). Validate in your sample system, as observed bands may shift due to post-translational modifications.
  • In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), consider optimizing antigen retrieval and fixation protocols. Reported cross-reactivity with mouse and rat suggests broad utility, but validation in your specific model is recommended.
  • ELISA can be used for antibody titering or screening purposes; follow standard protocols and include appropriate controls.
