Biological Background: TOM20 Function and Localization

  • TOM20 is a central receptor component of the translocase of the outer membrane of mitochondria (TOM) complex, essential for recognizing and translocating cytosolically synthesized mitochondrial preproteins.
    Related references:
    PMID:40080546
  • Together with TOMM22, it functions as the transit peptide receptor at the mitochondrial surface, facilitating the movement of preproteins into the TOM40 translocation pore.
    Related references:
    PMID:18331822
  • The TOM complex associates with VDAC2 and PINK1 kinase at depolarized mitochondria, stabilizing PINK1 at the outer membrane and triggering mitophagy via recruitment of the E3 ubiquitin ligase PRKN.
    Related references:
    PMID:40080546
  • TOM20 is also required for the translocation of cytochrome P450 monooxygenases across the mitochondrial outer membrane.
  • It is localized to the mitochondrion outer membrane.
  • The protein features a transmembrane helix and is subject to isopeptide bond formation, phosphorylation, and ubiquitination.

Experimental Guidance and Technical Tips

  • For Western blot (WB), consider loading whole cell lysates or mitochondrial fractions; a band at ~16 kDa is expected for TOM20.
  • For immunofluorescence (IF/ICC, IF-P), verify mitochondrial staining pattern in cultured cells; co-localization with mitochondrial markers can confirm specificity.
  • For immunohistochemistry (IHC-P), antigen retrieval may be required; test on tissues known to express TOM20 (e.g., heart, skeletal muscle).
  • This antibody is reported to react with human, mouse, and rat, enabling cross-species studies; validate in the relevant sample system.
  • The immunogen is a recombinant protein of human TOM20; the antibody is expected to recognize native protein in various applications.
