Biological Background: TOM20 Function and Localization
- Mitochondrial import receptor subunit TOM20 homolog, also designated MAS20, MOM19, or TOM20, is encoded by the TOMM20 gene.
- Central receptor component of the translocase of the outer membrane (TOM) complex; together with TOMM22, it recognizes and binds cytosolic mitochondrial preproteins via their N-terminal targeting sequences, directing them into the TOM40 translocation pore. Related references: PMID:40080546 PMID:18331822
- At depolarized mitochondria, the TOM complex associates with VDAC2 and PINK1 kinase; this interaction stabilizes PINK1 on the outer membrane and triggers mitophagy through the recruitment of the E3 ubiquitin ligase PRKN (Parkin). Related references: PMID:40080546
- Required for the translocation across the mitochondrial outer membrane of cytochrome P450 monooxygenases (by similarity).
- Subcellular localization: firmly anchored in the mitochondrial outer membrane via a single C-terminal transmembrane helix (residues 126–145), with the N-terminal domain exposed to the cytosol.
- Post-translational modifications include phosphorylation at multiple residues and isopeptide bonds (Ubl conjugation); these may regulate receptor activity and protein turnover.
- A key player in mitochondrial biogenesis, quality control, and cell survival, making it a valuable target in studies of neurodegeneration, metabolic disorders, and cancer.
Experimental Guidance and Technical Tips
- This antibody is pre-labeled with , a far-red fluorescent dye, permitting direct detection without secondary antibodies in immunofluorescence (IF/ICC, IF-P) and in western blotting (WB) when using near-infrared imaging systems.
- The immunogen encompasses amino acids 20–145 of human TOM20, covering the cytosolic receptor domain; enriching mitochondrial fractions or using organelle-specific markers can improve signal specificity.
- Demonstrates cross-reactivity with human, mouse, and rat TOM20; validate in the appropriate cell line (e.g., HeLa, HepG2, NIH/3T3) or tissue.
- For WB, expect a band migrating at approximately 16 kDa; for IF, co-staining with a mitochondrial marker (e.g., anti-COX IV, MitoTracker) confirms outer membrane localization.
- Optimization of antibody dilution is recommended; typical starting concentrations for conjugated antibodies range from 1:50 to 1:200 for IF and 1:500 to 1:2000 for WB, but must be titrated in the experimental system.
