Biological Background: TNRC6A Function and Localization
- TNRC6A, also known as GW182, TNRC6, CAGH26, or GW1, is a 210 kDa protein encoded by the TNRC6A gene on chromosome 8. It is a key component of the RNA-induced silencing complex (RISC).
- Functions in RNA-mediated gene silencing by microRNAs (miRNAs) and siRNAs. It is required for miRNA-dependent translational repression and siRNA-dependent endonucleolytic cleavage.
- Acts as a scaffolding protein, associating with argonaute proteins bound to partially complementary mRNAs, and recruiting the CCR4-NOT and PAN deadenylase complexes to facilitate mRNA decay.
- Localizes predominantly to the cytoplasm, specifically enriched in processing bodies (P-bodies), which are sites of mRNA degradation and storage.
- Ubiquitously expressed across human tissues, reflecting its fundamental role in post-transcriptional gene regulation.
- Post-translationally phosphorylated and undergoes alternative splicing, producing multiple isoforms that may modulate its activity or interactions.
- Contains RNA-binding domains and a coiled-coil region; involved in host-virus interactions and linked to epilepsy through genetic association studies.
Experimental Guidance and Technical Tips
- The immunogen used is a synthetic peptide corresponding to amino acids 950–1050 of human TNRC6A (NP_055309.2). This region may be conserved across species, supporting cross-reactivity with mouse and rat.
- For western blot (WB), consider using whole-cell lysates from human, mouse, or rat cell lines, as TNRC6A is ubiquitously expressed. The predicted molecular weight is ~210 kDa; ensure adequate gel resolution and transfer conditions for high-molecular-weight proteins.
- In immunofluorescence (IF) or immunocytochemistry (ICC), cytoplasmic and P-body localization is expected. Co-staining with P-body markers (e.g., DCP1A) may confirm specificity.
- For ELISA applications, validate the antibody using recombinant TNRC6A or peptide blocking assays to confirm binding affinity and specificity.
- As a polyclonal antibody, lot-to-lot variability may occur. Include appropriate controls and optimize dilution factors for each experimental setup.
