Biological Background: TNR Function and Localization
- Tenascin-R (TNR), also known as janusin or restrictin, is a secreted neural extracellular matrix protein belonging to the tenascin family.
- It mediates both adhesive and anti-adhesive interactions, capable of evoking stable adhesion and differentiation or repulsion and inhibition of neurite outgrowth depending on the cellular context.
- Binds to cell surface gangliosides to inhibit integrin-mediated cell adhesion and FAK phosphorylation; interaction with sulfatides promotes oligodendrocyte adhesion and differentiation.
- Interaction with contactin-1 (CNTN1) induces neuronal repulsion and inhibits neurite outgrowth; association with sodium channel subunit beta-2 (SCN2B) may cluster and regulate sodium channels at nodes of Ranvier.
- TNR-linked chondroitin sulfate glycosaminoglycans mediate binding to fibronectin (FN1), contributing to the inhibition of cell adhesion and neurite outgrowth.
- Localized to the extracellular space and matrix, with expression restricted to the brain.

- For Western blot (WB), the predicted molecular weight is approximately 150 kDa; note that isoforms and post-translational modifications (e.g., glycosylation, proteoglycan attachment) may produce additional bands.
- For ELISA, the antibody can be used to detect recombinant or native TNR; validate performance in the relevant sample matrix (e.g., brain tissue lysates or extracellular matrix preparations).
