Biological Background: Transportin-1 (TNPO1) Function and Localization
- Transportin-1 (importin beta-2 / KPNB2 / MIP) is a member of the karyopherin beta family, involved in nucleocytoplasmic transport.
- It functions as a nuclear import receptor for M9-containing cargo proteins, such as hnRNP A1 and A2, histones H2A, H2B, H3, H4, and ribosomal proteins.
- The protein localizes to both the cytoplasm and nucleus, shuttling between these compartments in a Ran-dependent manner.
- Mediates nuclear import of ribosomal proteins RPL23A, RPS7 and RPL5, and SRP19. Related references: PMID:11682607
- Mediates nuclear import of ADAR1 isoforms in a RanGTP-dependent fashion. Related references: PMID:19124606 PMID:24753571
- Serves as the main import receptor for BAP1, a component of the PR-DUB complex, with redundancy provided by KPNA1 and KPNA2. Related references: PMID:35446349
- Post-translational modifications include acetylation, and alternative splicing generates multiple isoforms.
- In HIV-1 infection, TNPO1 mediates nuclear import of the viral Rev protein, facilitating viral replication.
Experimental Guidance and Technical Tips
- The immunogen consists of a synthetic peptide from the C-terminal region (aa 790–890), likely recognizing full-length TNPO1; consider validating specificity in lysates from cells with known TNPO1 expression.
- For Western blot (WB), the predicted molecular weight is approximately 102 kDa; a 6–8% SDS-PAGE gel may be optimal for resolution.
- In ELISA applications, the peptide immunogen can serve as a positive control for assay optimization.
- Cross-reactivity is reported with human, mouse, and rat TNPO1; confirm reactivity in the relevant species by preliminary testing.
