Biological Background: TNFRSF6B Function and Localization
- Tumor necrosis factor receptor superfamily member 6B (TNFRSF6B), also known as Decoy receptor 3 (DcR3) or M68, is a secreted soluble receptor that lacks transmembrane and cytoplasmic domains.
- DcR3 functions as a decoy receptor by binding and neutralizing TNF family ligands, including TNFSF14/LIGHT, TNFSF15, and FASLG, thereby blocking apoptosis and other immune-signaling pathways mediated by their cognate receptors.
- The protein is characterized by disulfide bond-stabilized cysteine-rich repeats and is glycosylated, typical of TNF receptor family members.
- Subcellular localization: Secreted into the extracellular space.
- Highly expressed in several primary tumors from colon, stomach, rectum, and esophagus, and in SW480 colon carcinoma cells, suggesting a role in immune evasion in cancer.
- Detected in normal adult tissues including stomach, spinal cord, lymph node, trachea, spleen, colon, and lung, and in fetal lung, brain, and liver.
- Keywords: 3D-structure, Apoptosis, Disulfide bond, Glycoprotein, Receptor, Repeat, Secreted, Signal.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a region within the extracellular domain of TNFRSF6B (amino acids 200-300), which is expected to yield antibodies recognizing the native secreted protein.
- Suitable for Western blotting (WB) and ELISA applications; optimal dilutions and conditions should be empirically determined.
- For WB, consider using the predicted molecular weight of 33 kDa as a reference.
- Cross-reactivity confirmed with human and rat; validate in the relevant sample system.
