  • TNFRSF11B encodes osteoprotegerin (OPG), also known as osteoclastogenesis inhibitory factor (OCIF), a secreted glycoprotein belonging to the tumor necrosis factor receptor superfamily.
  • Acts as a decoy receptor for TNFSF11/RANKL, neutralizing its function and thereby inhibiting osteoclast differentiation and activation.
  • Plays a critical role in bone homeostasis; the local ratio of RANKL to OPG determines the extent of bone resorption.
  • May also function as a decoy receptor for TNFSF10/TRAIL, blocking TRAIL-induced apoptosis and further inhibiting osteoclastogenesis.
