Biological Background: TNFRSF10D/TRAILR4 Function and Localization

  • Tumor necrosis factor receptor superfamily member 10D (TNFRSF10D), also known as Decoy receptor 2 (DcR2), TRAIL receptor 4 (TRAIL-R4), CD264, and TRAIL receptor with a truncated death domain.
  • Functions as a receptor for the cytotoxic ligand TRAIL. Due to its truncated death domain, it does not induce apoptosis but instead protects cells against TRAIL-mediated apoptosis. Related references: PMID:9430226, PMID:9537512
  • The receptor’s ability to induce the NF-kB pathway is controversial: some studies report it cannot (PubMed:9382840), while others suggest it can (PubMed:9430226). Related references: PMID:9382840, PMID:9430226
  • Localized to the cell membrane as a transmembrane receptor.
  • Widely expressed in tissues, with high levels in fetal kidney, lung, liver, adult testis and liver, and also present in peripheral blood leukocytes, colon, small intestine, ovary, prostate, thymus, spleen, pancreas, kidney, lung, placenta, and heart.
  • Contains a signal peptide, extracellular cysteine-rich repeats, a transmembrane helix, and a truncated cytoplasmic death domain; it is a glycoprotein with disulfide bonds.
  • Encoded by the TNFRSF10D gene (Gene ID: 8793), belonging to the tumor necrosis factor receptor superfamily, which plays a role in apoptosis regulation.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 56-211, which lies within the extracellular domain; therefore, the antibody is likely to recognize the native surface receptor.
  • Validated for flow cytometry (FC); appropriate fixation and permeabilization conditions should be optimized for intracellular staining if needed.
  • As a monoclonal antibody, it is expected to exhibit high specificity; cross-reactivity with other TRAIL receptors should be assessed if necessary.
  • Consider using this antibody in functional assays to study TRAIL-induced signaling or in combination with other markers to characterize cell populations.
