  • Tumor necrosis factor receptor superfamily member 1B (TNFR2/TNFRSF1B), also known as p75, CD120b, or TNF-R-II, is encoded by the TNFRSF1B gene in humans.
  • TNFR2 functions as a high-affinity receptor for tumor necrosis factor (TNF/TNFSF2) and has approximately 5-fold lower affinity for lymphotoxin-alpha (TNFSF1).
  • The receptor mediates most of the metabolic effects of TNF, and its cytoplasmic domain recruits the TRAF1/TRAF2 complex, which in turn recruits the apoptotic suppressors BIRC2 and BIRC3 to modulate apoptotic signaling.
  • An alternatively spliced isoform (isoform 2) acts as a dominant-negative regulator by blocking TNF-induced apoptosis, suggesting a role in fine-tuning TNF responses.
