Biological Background: TNFAIP8L2 Function and Localization
- TNFAIP8L2 (Tumor necrosis factor alpha-induced protein 8-like protein 2, also known as TIPE2 or Inflammation factor protein 20) is a member of the TNFAIP8 family and is characterized as a phosphoprotein with ubiquitin-like conjugation (Ubl conjugation) potential.
- Acts as a negative regulator of innate and adaptive immunity by maintaining immune homeostasis. Related references: PMID:27043859
- Modulates the Toll-like receptor (TLR) signaling pathway, determining the strength of lipopolysaccharide (LPS)-induced signaling and gene expression. Related references: PMID:32188758
- Inhibits T-cell receptor (TCR)-mediated T-cell activation, thereby negatively regulating T-cell function to prevent hyperresponsiveness (By similarity).
- Inhibits autolysosome formation by negatively modulating MTOR activation; interacts with RAC1 to promote disassociation of the RAC1-MTOR complex. Related references: PMID:32460619
- Serves as a checkpoint in natural killer (NK)-cell biology, correlating with NK-cell maturation and negatively regulating NK-cell maturation and antitumor immunity; suppresses IL-15-triggered mTOR activity in NK-cells (By similarity).
- Localizes to the cytoplasm, nucleus, and lysosome.
- Expressed in immune cells (T-cells, B-cells, macrophages) as well as neurons in the brain and brainstem, and in stratified squamous epithelia of the esophagus, cervix, and skin.
Experimental Guidance and Technical Tips
- The recombinant immunogen corresponds to amino acids 1-184 of human TNFAIP8L2, covering the N-terminal portion of the protein; this region may be suitable for detecting total TNFAIP8L2. Verify reactivity with relevant post-translational modifications if needed.
- For western blotting (WB): The predicted molecular weight is 21 kDa; consider using a 10-15% SDS-PAGE gel for optimal resolution. Validate in lysates from human, mouse, or rat cell lines/tissues with appropriate controls.
- For immunofluorescence/immunocytochemistry (IF/ICC): Expect cytoplasmic, nuclear, and lysosomal staining. Optimize fixation, permeabilization, and co-localization markers for your specific system.
- For immunoprecipitation (IP) and ELISA: Use standard protocols with protein A/G agarose for IP; for ELISA, titrate antibody concentration and consider using recombinant protein or peptide coating for validation.
