Biological Background: TNFAIP6 Function and Localization

  • TNFAIP6 (Tumor necrosis factor-inducible gene 6 protein), also known as TSG-6, is a secreted protein that acts as a major regulator of extracellular matrix organization during tissue remodeling. It catalyzes the transfer of heavy chains from inter-alpha-inhibitor to hyaluronan, potentiating the antiprotease function of I-alpha-I complex. Related references: PMID:15917224 | PMID:16873769 | PMID:20463016
  • Essential for hyaluronan-rich matrix formation around the cumulus cell-oocyte complex, a critical step for oocyte fertilization. Related references: PMID:26468290
  • Assembles hyaluronan in pericellular matrices to facilitate receptor clustering and signaling. Promotes macrophage extravasation by enabling hyaluronan binding to LYVE1 on lymphatic endothelium and switches CD44 on vascular endothelium into an active state to support leukocyte rolling. Related references: PMID:15060082 | PMID:26823460
  • Modulates chemokine interactions with extracellular matrix and endothelial proteoglycans, potentially disrupting chemokine gradient formation. Limits excessive neutrophil recruitment by antagonizing CXCL8 binding to glycosaminoglycans on vascular endothelium. Related references: PMID:27044744 | PMID:24501198
  • Plays a role in osteogenesis and bone remodeling: inhibits BMP2-dependent differentiation of mesenchymal stem cells into osteoblasts and protects against inflammatory bone erosion by suppressing RANKL-dependent osteoclast activation. Related references: PMID:16771708 | PMID:18586671
  • Localized to the extracellular space (secreted).
  • Tissue expression: Found in airway epithelium, submucosal glands, and bronchoalveolar lavage fluid. Also expressed in mesenchymal stem cells and present in synovial fluid of rheumatoid arthritis patients. Related references: PMID:16873769 | PMID:16771708
  • Keywords include calcium binding, cell adhesion, glycoprotein, hydrolase, and disulfide bond, reflecting its role in matrix organization and inflammation.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to nearly full-length human TNFAIP6 (aa 18–277), so the antibody may recognize both native and recombinant protein. Consider using this antibody for detection of TNFAIP6 in conditioned media or serum samples where the protein is secreted.
  • For WB, a band at approximately 31 kDa is expected. Since TNFAIP6 is a glycoprotein, some shift or additional bands may be observed in certain cell types; validate in the relevant sample system.
  • Cross-reactivity with mouse and rat is reported, but sequence alignment suggests potential reactivity with other species; experimental validation is recommended if used outside human/mouse/rat.
  • For ELISA, this polyclonal antibody can be used as a capture or detection reagent after pairing with a monoclonal or other polyclonal antibody targeting distinct epitopes. Optimization of coating and detection conditions is advised.
  • As a secreted protein, TNFAIP6 may be present at low levels in serum; enrichment steps or high-sensitivity detection methods might be needed for some applications.
