Biological Background: TNF-alpha Function and Localization
- Tumor necrosis factor (TNF-alpha, cachectin, TNFSF2) is a key pro-inflammatory cytokine belonging to the tumor necrosis factor ligand superfamily.
- Mainly secreted by macrophages, TNF-alpha binds to TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR, inducing cell death in certain tumor cell lines and acting as a potent pyrogen.
- Under certain conditions, TNF-alpha can stimulate cell proliferation and differentiation, and plays a role in angiogenesis by synergistically inducing VEGF with IL1B and IL6.
- TNF-alpha induces insulin resistance in adipocytes via inhibition of IRS1 tyrosine phosphorylation and glucose uptake, partially mediated by GKAP42 protein degradation. Related references: PMID:25586176
- It promotes osteoclastogenesis and bone resorption. Related references: PMID:32741026
- The TNF intracellular domain (ICD) form induces IL12 production in dendritic cells.
- Subcellularly, TNF-alpha is found at the cell membrane and is also secreted; it contains a signal-anchor, transmembrane helix, and undergoes post-translational modifications including glycosylation, disulfide bonds, and phosphorylation.
Experimental Guidance and Technical Tips
- The immunogen (aa 80-235) targets the extracellular domain of mouse TNF-α, covering the receptor-binding region; it is expected to recognize both the membrane-bound and secreted soluble forms.
- For Western blotting, a major band around 26 kDa may be observed; additional bands or shifts could arise from glycosylation or proteolytic processing—validate using appropriate controls.
- In immunofluorescence/immunocytochemistry, consider permeabilization protocols to balance detection of intracellular and membrane-anchored TNF—signal may appear in the Golgi, plasma membrane, or as secreted puncta depending on cell type and stimulation.
- For ELISA, antibody pairing compatibility should be assessed if used as a capture/detection antibody in sandwich assays; cross-reactivity with other TNF superfamily members is unlikely based on the immunogen design but should be confirmed.
