Biological Background: TMEM43 Function and Localization

  • Encoded by the TMEM43 gene, transmembrane protein 43 (also known as LUMA) is a multi-pass membrane protein of approximately 45 kDa (calculated MW 44876 Da) with diverse roles in nuclear envelope architecture, innate immunity, and cellular signaling.
  • Plays a critical role in maintaining nuclear envelope structure by organizing protein complexes at the inner nuclear membrane and is required for retaining emerin (by similarity).
  • Modulates innate immune signaling through the cGAS-STING pathway by interacting with RNF26, linking nuclear envelope integrity to antiviral responses. Related references: PMID:32614325
  • Functions as a signaling adapter mediating NF-kappa-B activation downstream of EGFR and upstream of CARD10, implicating TMEM43 in inflammatory and proliferative pathways. Related references: PMID:27991920

Experimental Guidance and Technical Tips

  • For Western blot, expect a specific band at approximately 45 kDa. Positive controls may include human placental tissue lysate or cell lines known to express TMEM43; antibody reactivity has been confirmed for human, mouse, and rat samples.
  • In paraffin-embedded immunohistochemistry (IHC-P), antigen retrieval is typically required. Optimal staining may be observed in tissues with high endogenous TMEM43 levels, such as placenta; however, expression in heart, ovary, and other tissues can also be informative after in-house validation.
  • For ELISA applications, the recombinant immunogen (aa 35–135) serves as a suitable reference standard. Users should establish assay conditions and linear range through titration experiments.
  • Given the monoclonal nature of this antibody, consistent lot-to-lot performance is expected. However, as with any antibody, validation in the specific experimental system is recommended.
