Biological Background: TMEM2 Function and Localization
- TMEM2, also known as CEMIP2 or Cell migration-inducing hyaluronidase 2, is a cell surface hyaluronidase encoded by the CEMIP2 gene, with a molecular weight of approximately 154 kDa.
- It specifically mediates the initial cleavage of high-molecular-weight hyaluronan into intermediate fragments of about 10-5 kDa, and does not act on chondroitin sulfate or dermatan sulfate. Related references: PMID:37527776
- TMEM2 plays an essential role in systemic hyaluronan catabolism and turnover, regulating cell adhesion and migration by degrading hyaluronan at focal adhesion sites (By similarity).
- It acts as a regulator of angiogenesis and heart morphogenesis by mediating degradation of extracellular hyaluronan, thereby modulating VEGF signaling (By similarity).
- The protein localizes to the cell membrane and is widely expressed across tissues.
- Key structural features include a signal-anchor, transmembrane domain, and lectin-like repeats; it undergoes post-translational modifications such as glycosylation and phosphorylation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a peptide within the C-terminal region (amino acids 1150-1250) of human TMEM2. This region may be suitable for detecting both human and the cross-reactive species mouse and rat, but sequence alignment is recommended to confirm epitope conservation.
- For Western blot, a band around 154 kDa is expected. Consider using appropriate gel percentages and transfer conditions for high-molecular-weight proteins.
- ELISA may be used for detecting TMEM2 in cell lysates or purified protein; optimize coating and detection conditions as per standard protocols.
- Since TMEM2 is a transmembrane protein, sample preparation should include membrane fraction enrichment for optimal detection.
