- TMC1 is a pore-forming subunit of the mechanotransducer (MET) non-selective cation channel complex, essential for auditory sensory transduction.
- The MET channel is located at the tips of stereocilia of cochlear hair cells and is activated by tension in the tip-link extending from one stereocilium to the adjacent shorter stereocilium.
- TMC1 forms dimeric ion-conducting subunits within the MET complex, aided by auxiliary proteins LHFPL5, TMIE, CIB2/3, TOMT, and PCDH15.
- TMC1 MET channel is highly permeable to calcium and likely transports monovalent cations (By similarity).
- It is also involved in vestibular hair cells transduction current (By similarity).
- Subcellularly, TMC1 localizes to the cell membrane.
- Post-translational modifications: TMC1 is classified as a phosphoprotein.
- Tissue specificity: Detected in fetal cochlea and at low levels in placenta and testis.
