Biological Background: TM7SF2 Function and Localization
- TM7SF2 (Transmembrane 7 superfamily member 2) is also known as Delta(14)-sterol reductase, ANG1, C14SR, NET47, and DHCR14A. It belongs to the sterol reductase family.
- It catalyzes the reduction of the C14-unsaturated bond of lanosterol, a critical step in the cholesterol biosynthesis pathway.
- TM7SF2 is a multi-pass membrane protein localized primarily to the endoplasmic reticulum membrane and microsome membrane.
- The gene exhibits alternative splicing, yielding multiple transcript variants; the canonical isoform has a predicted molecular weight of approximately 46 kDa.
- It is widely expressed in adult tissues, including heart, brain, pancreas, lung, liver, skeletal muscle, kidney, ovary, prostate, testis, and adrenal gland, but is absent in placenta, spleen, thymus, small intestine, colon mucosa, and peripheral blood leukocytes.
- As an oxidoreductase that acts on the CH-CH group of donors with NAD or NADP as acceptor, TM7SF2 is involved in steroid and sterol metabolic processes.
Experimental Guidance and Technical Tips
- The immunogen used to generate this antibody corresponds to amino acids 160-250 of human TM7SF2, a region within the central portion of the protein. This sequence is conserved between human and mouse, supporting cross-reactivity.
- This antibody is suitable for Western blot (WB) and ELISA applications. For WB, consider a starting dilution range based on your sample type and detection system, and validate with appropriate controls.
- Due to the transmembrane nature of TM7SF2, ensure adequate solubilization of membrane fractions during sample preparation. The predicted band size is ~46 kDa.
- For ELISA, test against recombinant protein or cell lysates expressing TM7SF2 to assess binding specificity.
