Biological Background: TLR6 Function and Localization

  • TLR6 (Toll-like receptor 6) is a member of the Toll-like receptor (TLR) family, which plays a fundamental role in pathogen recognition and activation of innate immunity. It is a single-pass type I membrane protein containing an extracellular domain with leucine-rich repeats (LRR), a transmembrane domain, and a cytoplasmic Toll/interleukin-1 receptor (TIR) domain.
  • TLR6 specifically recognizes diacylated and, to a lesser extent, triacylated lipopeptides from Gram-positive bacteria and fungi. It forms an activation cluster with TLR2, CD14, and CD36 at the cell surface, triggering signaling from membrane rafts and subsequent Golgi targeting. Related references: PMID:16880211 PMID:20037584
  • Signaling downstream of TLR2:TLR6 is mediated via MYD88 and TRAF6, leading to NF-kappa-B activation, cytokine secretion, and the inflammatory response. Cooperatively with TLR2, it recognizes microbial components such as mycoplasmal MALP-2, soluble tuberculosis factor (STF), phenol-soluble modulin (PSM), and B. burgdorferi OspA-L.   • In a distinct pathway, TLR6 can complex with TLR4 to promote sterile inflammation in monocytes/macrophages in response to oxidized low-density lipoprotein (oxLDL) or amyloid-beta 42, with CD36 serving as the initial sensor.   • Subcellularly, TLR6 is located at the cell membrane, within cytoplasmic vesicles, on the phagosome membrane, in membrane rafts, and at the Golgi apparatus, reflecting its dynamic trafficking upon activation.
  • The protein undergoes post-translational modifications including glycosylation and disulfide bond formation, consistent with its presence in the secretory pathway and on the cell surface.
  • TLR6 is expressed in monocytes, CD11c+ immature dendritic cells, plasmacytoid pre-dendritic cells, and dermal microvessel endothelial cells, indicating its role in immune surveillance in barrier tissues.
  • Alternative splicing generates multiple isoforms, which may modulate ligand specificity or signaling outputs.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide from the C-terminal region (aa 697–796) of human TLR6, which likely corresponds to the cytoplasmic domain. This design favors recognition of total TLR6 irrespective of glycosylation or ectodomain shedding.
  • For Western blotting, the predicted molecular weight is ~92 kDa, but the mature glycosylated form often migrates at a higher apparent molecular weight. Use reducing conditions and validate band specificity with appropriate controls.
  • In immunofluorescence/immunocytochemistry (IF/ICC), expect plasma membrane and intracellular vesicular staining. Detergent permeabilization is required for detection of intracellular pools. Co-staining with organelle markers can reveal activation-induced trafficking.
  • The antibody cross-reacts with human and mouse TLR6; verify reactivity in the desired species and tissue by positive and negative controls.
  • ELISA applications may be developed using the polyclonal antibody as a capture or detection reagent; consider pairing with a monoclonal antibody for enhanced specificity.
