Biological Background: TLR3 Function and Localization

  • Protein identity: Toll-like receptor 3 (TLR3), also known as CD283, is a member of the Toll-like receptor (TLR) family. It is encoded by the TLR3 gene (GeneID: 7098) and exists in alternatively spliced isoforms.
  • Core function: Serves as a key component of innate and adaptive immunity by recognizing double-stranded RNA (dsRNA), a molecular signature of viral infection, leading to NF-κB activation and IRF3 nuclear translocation via the TRIF/TICAM1 adapter.
  • Gene family: Belongs to the conserved Toll-like receptor family, which includes pattern recognition receptors that detect pathogen-associated molecular patterns.
  • Subcellular localization: Localizes to the endoplasmic reticulum membrane, endosome membrane, and early endosome, consistent with its role in sensing internalized viral RNA.
  • Post-translational modifications and structural features: Contains leucine-rich repeats, a signal sequence, and transmembrane helix; is modified by glycosylation, disulfide bond formation, phosphorylation, and ubiquitin-like conjugation.
  • Tissue specificity: Highly expressed in placenta and pancreas, and in CD11c+ immature dendritic cells. It is the TLR most strongly expressed in the brain, including astrocytes, glia, and neurons.
  • Antimicrobial and signaling roles: Upon dsRNA binding, triggers inflammatory responses, cytokine secretion, and antiviral immunity, highlighting its critical role in host defense.

Experimental Guidance and Technical Tips

  • The immunogen was designed using amino acids 500–700 of human TLR3, which maps to the extracellular leucine-rich repeat domain implicated in dsRNA binding. This design may enhance detection of the native, full-length receptor.
  • For Western blotting, the predicted molecular weight is 104 kDa; however, due to glycosylation and other modifications, the observed band may appear slightly larger. Validate the antibody in your specific sample system.
  • In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), consider testing both frozen and paraffin-embedded sections, and optimize antigen retrieval conditions as needed.
  • The antibody cross-reacts with mouse and rat TLR3, making it suitable for studies in these common model organisms, but confirmation of reactivity in your specific experimental context is recommended.
  • For ELISA applications, the polyclonal nature may provide enhanced sensitivity through recognition of multiple epitopes; however, use appropriate controls to ensure specificity.
