Biological Background: TLR2 Function and Localization

- Acts via MYD88 and TRAF6, leading to NF-kappa-B activation, cytokine secretion, and the inflammatory response.

Experimental Guidance and Technical Tips
- The immunogen corresponds to the intracellular TIR domain (aa 636–784 of human TLR2), a region conserved across species, supporting the reported cross-reactivity with mouse and rat.
- For Western blot (WB), expect a band at ~90 kDa. Use appropriate positive controls (e.g., THP-1 or PBMC lysates) and consider validating specificity via blocking peptides or siRNA knockdown.
- In immunofluorescence/immunocytochemistry (IF/ICC), permeabilization is required to detect intracellular TLR2. Optimize fixation and antibody dilution for your cell type.
- The antibody has been used in ELISA; when developing sandwich ELISAs, pairing with a capture antibody against a different epitope may improve sensitivity.
