Biological Background: TLK1 Function and Localization
- TLK1 (Tousled-like kinase 1), also known as PKU-beta, is a serine/threonine-protein kinase.
- It is rapidly and transiently inhibited by phosphorylation following DNA double-stranded breaks during S-phase, in a cell cycle checkpoint and ATM-pathway dependent manner, regulating chromatin assembly.
- In vitro, TLK1 phosphorylates histone H3 at Ser-10, suggesting a role in chromatin modification.
- Isoform 3 phosphorylates and stabilizes the t-SNARE SNAP23, enhancing its assembly with syntaxin, and also facilitates repair of DNA double-stranded breaks.
- TLK1 is primarily localized in the nucleus.
- It is widely expressed, present in fetal placenta, liver, kidney, and pancreas, but not in heart or skeletal muscle; isoform 3 is ubiquitously expressed.
