Biological Background: TLE4 Function and Localization

• TLE4 is a member of the transducin-like enhancer of split (TLE) family of transcriptional corepressors, also known as Grg-4 (Groucho-related protein 4).
• It functions as a transcriptional corepressor that binds to various transcription factors, inhibiting transcriptional activation mediated by PAX5 and by CTNNB1/TCF in Wnt signaling.
• The protein is essential for SIX3 transcriptional repressor activity during retina and lens development, and also mediates MSX1-dependent repression of CGA/alpha-GSU.
• TLE4 localizes to the nucleus and is expressed in most tissues, with highest levels in brain and muscle.
• Post-translational modifications include acetylation and phosphorylation; the protein contains WD repeats and is subject to ubiquitination.
