Biological Background: TLE1 Function and Localization
- TLE1, also known as Transducin-like enhancer protein 1 or enhancer of split groucho-like protein 1 (ESG1), is a member of the Groucho/TLE family of transcriptional corepressors.
- It functions as a transcriptional corepressor that binds to a variety of transcription factors, inhibiting NF-kappa-B-regulated gene expression, and repressing the transcriptional activation mediated by FOXA2 and by CTNNB1/TCF in the Wnt signaling pathway.
- Unusually, TLE1 can also act as a transcriptional coactivator for the nuclear receptor ESRRG, highlighting its context-dependent regulatory roles.
- The repressive effects of full-length TLE family members may be modulated through association with the dominant-negative protein AES.
- TLE1 is predominantly localized in the nucleus, where it mediates its transcriptional regulatory functions.
- Expression is detected in all tissues examined, with highest levels observed in the brain, liver, and muscle.
- The protein features multiple WD repeat domains and is subject to post-translational modifications including phosphorylation and ubiquitin-like conjugation, as indicated by UniProt keywords.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from the internal region (aa 150–250) of human TLE1; consider using a blocking peptide to confirm antibody specificity in your assay.
- For Western blotting, TLE1 has a predicted molecular weight of ~83 kDa; ensure adequate separation and transfer conditions, and consider nuclear-enriched lysates if the signal is weak.
- In immunohistochemistry (IHC-P), validate staining with appropriate positive (e.g., brain or liver tissue) and negative controls, as TLE1 is widely expressed but may show tissue-specific patterns.
- ELISA applications may be optimized by titrating the antibody against the immunogen peptide or recombinant TLE1 protein to establish sensitivity and dynamic range.
- Given the cross-reactivity with mouse and rat, the antibody can be used for interspecies studies, but validate specificity independently in each species due to potential sequence variations.
