- TIR1 (TRANSPORT INHIBITOR RESPONSE 1), also known as weak ethylene-insensitive protein 1 (WEI1), is an auxin receptor that functions as the substrate recognition component of the SCF(TIR1) E3 ubiquitin ligase complex.
- It mediates the ubiquitin-dependent proteasomal degradation of Aux/IAA transcriptional repressors, thereby regulating auxin-responsive gene transcription.
- TIR1 plays a central role in auxin-mediated developmental processes including root and hypocotyl growth, lateral root formation, cell elongation, and gravitropism.
- The protein is localized to the nucleus, consistent with its role in transcriptional regulation.
- It is broadly expressed in roots, stems, leaves, and flowers, with strong expression in floral stigma, anther filaments, abscission zones, and vascular tissues of adult plants.
- Beyond auxin signaling, TIR1 also participates in ethylene signaling in roots and contributes to plant defense by conferring sensitivity to the bacterial pathogen Pseudomonas syringae.
