Biological Background: TIN2/TINF2 Function and Localization
- TIN2, also known as TERF1-interacting nuclear factor 2 or TRF1-interacting nuclear protein 2, is encoded by the TINF2 gene. Alternative splicing produces isoforms 1 and 2.
- TIN2 is a core component of the shelterin (telosome) complex, which binds double-stranded telomeric TTAGGG repeats to safeguard chromosome ends.
- The shelterin complex protects telomeres from being recognized as DNA damage, preventing inappropriate activation of DNA repair pathways at chromosome termini.
- TIN2 facilitates the assembly of the shelterin complex and may contribute to telomere length regulation and protection.
- Isoform 1 has been proposed to play an additional role in tethering telomeres to the nuclear matrix.
- Subcellular localization: TIN2 is primarily found in the nucleus, at telomeres, and possibly associated with the nuclear matrix.
- Tissue expression: Detected in heart, brain, placenta, lung, liver, skeletal muscle, kidney, and pancreas.
- TIN2 undergoes post-translational modifications including acetylation, phosphorylation, and ubiquitin conjugation (isopeptide bond). Mutations in TINF2 are associated with dyskeratosis congenita and related telomere biology disorders.

- The immunogen comprises amino acids 210–309 of human TIN2/TINF2; confirm sequence conservation across human, mouse, and rat to support cross-reactivity expectations.
- For Western blotting, consider using whole-cell lysates or nuclear extracts from human, mouse, or rat cell lines; the predicted molecular weight is ~50 kDa (UniProt: 50023 Da).
- Validate the antibody in the relevant sample system and include appropriate positive and negative controls, such as siRNA-mediated knockdown lysates or known TIN2-expressing cell lines.
