Biological Background: TIMP3 Function and Localization
- TIMP3 is a member of the tissue inhibitor of metalloproteinases (TIMP) family, playing a critical role in extracellular matrix (ECM) regulation and turnover.
- It reversibly inhibits a broad spectrum of matrix metalloproteinases (MMPs), including MMP-1, MMP-2, MMP-3, MMP-7, MMP-9, MMP-13, MMP-14, and MMP-15, thereby modulating ECM remodeling.
- TIMP3 also inhibits the alpha-secretases ADAM10 and ADAM17, regulating the processing of amyloid precursor protein (APP) and apolipoprotein E receptor ApoER2. Related references: PMID:17913923
- Functions as a tumor suppressor and potent angiogenesis inhibitor by directly interacting with VEGFR2/KDR, blocking VEGFA binding. Related references: PMID:12652295
- Selectively induces apoptosis in angiogenic endothelial cells through a caspase-independent pathway, involving disruption of focal adhesion kinase signaling. Related references: PMID:25558000
- The protein is secreted and localizes to the extracellular matrix, consistent with its role in ECM modulation.
- Contains multiple disulfide bonds and zinc-binding sites essential for its structural stability and inhibitory function.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide corresponding to amino acids 110-210 of human TIMP3 (NP_000353.1). Consider validating antibody specificity in your target species using peptide blocking or knockout models.
- For Western blot (WB), a band at approximately 24 kDa is expected in samples from human, mouse, or rat tissues known to express TIMP3.
- This polyclonal antibody is suitable for ELISA; optimal dilution and conditions should be determined empirically.
