- TIMP1 (Metalloproteinase inhibitor 1) is a secreted protein that functions as a key inhibitor of matrix metalloproteinases (MMPs), including collagenases, by forming 1:1 complexes and irreversibly inactivating them through binding to their catalytic zinc cofactor.
- It acts on a broad range of MMPs such as MMP1, MMP2, MMP3, MMP7, MMP8, MMP9, MMP10, MMP11, MMP12, MMP13, and MMP16, but does not inhibit MMP14.
- Beyond MMP inhibition, TIMP1 acts as a growth factor that regulates cell differentiation, migration, and apoptosis, activating cellular signaling cascades via receptors CD63 and ITGB1.
- TIMP1 mediates erythropoiesis in vitro, stimulating the growth and differentiation of human and murine erythroid progenitors in a species-specific manner.
- The protein is also known as Erythroid-potentiating activity (EPA) and Fibroblast collagenase inhibitor, reflecting its dual roles.
