Biological Background: TIMM8B Function and Localization
- TIMM8B (Translocase of Inner Mitochondrial Membrane 8B), also designated DDP2 or TIM8B, is a mitochondrial import inner membrane translocase subunit Tim8 B.
- It functions as a probable mitochondrial intermembrane chaperone that participates in the import and insertion of multi-pass transmembrane proteins into the mitochondrial inner membrane, and assists in the transfer of β-barrel precursors from the TOM complex to the SAM complex.
- TIMM8B is thought to prevent aggregation of hydrophobic precursors and guide them through the intermembrane space (by similarity).
- Subcellularly, it localizes to the mitochondrion inner membrane.
- The protein is ubiquitously expressed, with highest levels in heart, kidney, liver, and skeletal muscle.
- Post-translational modifications include acetylation and formation of disulfide bonds; it also binds zinc, classifying it as a metal-binding chaperone.
- This small ~9 kDa protein is essential for mitochondrial protein transport and homeostasis, and is part of the small Tim protein family in the intermembrane space.

- The immunogen corresponds to amino acids 16–98 of human TIMM8B; for Western blot, expect a band near 9 kDa and use high-percentage gels with appropriate molecular weight markers.
- For IHC-P and IF/ICC, include antigen retrieval and permeabilization steps, and expect a mitochondrial staining pattern.
