Biological Background: TIMM8A Function and Localization

  • TIMM8A (Translocase of Inner Mitochondrial Membrane 8A), also known as DDP1, is a small ~11 kDa protein located in the mitochondrial intermembrane space and inner membrane.
  • Functions as a chaperone that facilitates the import and assembly of multi-pass transmembrane proteins into the mitochondrial inner membrane, including TIMM23 and the SLC25A12/A13 carriers. Also assists in transferring beta-barrel precursors from the TOM complex to the SAM complex.
  • Forms a heterohexameric complex with TIMM13 to protect hydrophobic precursors from aggregation and guide them through the intermembrane space.
  • Mutations in TIMM8A cause Mohr-Tranebjaerg syndrome (deafness-dystonia syndrome), highlighting its essential role in neurological development.
  • Contains conserved cysteine residues that form intramolecular disulfide bonds and binds zinc ions, critical for structural stability and chaperone activity.
  • Highly expressed in fetal and adult brain, with additional expression in heart, liver, kidney, and skeletal muscle.

Experimental Guidance and Technical Tips

  • For Western blot, use mitochondrial-enriched lysates to enhance detection of the 11 kDa band; a 12–15% Tris-glycine gel with low molecular weight markers is recommended.
  • In immunofluorescence/ICC, permeabilize cells to allow antibody access to the mitochondrial inner membrane; co-stain with a mitochondrial marker for colocalization.
  • The immunogen (aa 1–97) covers the full-length processed protein, so the antibody likely detects both monomeric and complexed TIMM8A.
  • Validate species reactivity using positive control lysates from human, mouse, or rat, as indicated by the cross-reactivity data.
  • For ELISA, use recombinant TIMM8A protein or peptide as coating antigen; cross-reactivity with closely related family members such as TIMM8B should be tested.
  • Avoid excessive reducing conditions if the disulfide-bonded epitope is critical for recognition.
