Biological Background: Timeless Function and Localization
- Protein timeless homolog, also known as TIM, TIM1, hTIM, or FASPS4, encoded by the TIMELESS gene; alternative splicing yields multiple isoforms.
- Plays an essential role in DNA replication and genome maintenance: forms a complex with TIPIN to stabilize replication forks, regulates CHEK1 phosphorylation, and activates the intra-S phase checkpoint in response to genotoxic stress; specifically required for the ATR-CHEK1 pathway upon hydroxyurea or UV treatment. Related references: PMID:17141802, PMID:17296725, PMID:23359676, PMID:15798197
- Promotes processive DNA replication past guanine-rich G-quadruplex (G4) structures, likely cooperating with DDX11. Related references: PMID:32705708
- Involved in DNA repair: accumulates at double-strand breaks and facilitates homologous recombination through interaction with PARP1. Related references: PMID:26344098, PMID:30356214, PMID:31138685
- Regulates circadian clock: negatively modulates CLOCK/BMAL1-mediated transactivation of PER1, may destabilize the PER2-CRY2 complex, and mediates DNA damage-dependent phase shifting. Related references: PMID:23418588, PMID:31138685, PMID:9856465
- Subcellular localization: predominantly nuclear, chromosome-associated.
- Tissue specificity: ubiquitously expressed across human tissues, with highest levels in placenta, pancreas, thymus, and testis.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the C-terminal region (aa 1058-1208) of human Timeless; consider epitope accessibility in your assay design.
- For Western blot, a low-percentage gel (e.g., 6–8%) is recommended to resolve the 139 kDa protein; validate transfer conditions for high-molecular-weight targets.
- In ELISA applications, optimize coating buffers and antibody concentrations; this monoclonal antibody may serve as a detection or capture reagent.
- Cross-reactivity has been confirmed for human samples; if using non-human species, validate specificity independently.
