Biological Background: TIM-3/HAVCR2 Function and Localization

  • HAVCR2/TIM-3 is generally accepted as an inhibitory receptor that regulates innate and adaptive immune responses, with context-dependent activity. It regulates macrophage activation. Related references: PMID:24825777, PMID:11823861
  • It inhibits Th1-mediated auto- and alloimmune responses and promotes immunological tolerance. On CD8+ T-cells, it attenuates TCR signaling by blocking NF-kappaB and NFAT, reducing IL-2 secretion, possibly via association with LCK. Related references: PMID:14556005, PMID:24337741, PMID:26492563
  • TIM-3 is a receptor for LGALS9, leading to suppression of T-cell responses and apoptosis of antigen-specific cells, though this role has been challenged. Related references: PMID:16286920, PMID:24337741, PMID:23555261
  • It also functions as a receptor for phosphatidylserine, mediating phagocytosis of apoptotic cells and positively regulating innate immunity in dendritic cells.
  • On natural killer (NK) cells, TIM-3 acts as a coreceptor enhancing IFN-gamma production in response to LGALS9, but can also suppress NK cell-mediated cytotoxicity. Related references: PMID:22323453, PMID:22383801
  • Expressed in Th1, Treg cells, dendritic cells, NK cells, and epithelial tissues; localized at the cell membrane and cell junctions.
  • TIM-3 is a transmembrane glycoprotein with immunoglobulin-like domains, disulfide bonds, and palmitoylation, with a predicted molecular weight of ~33 kDa.

Experimental Guidance and Technical Tips

  • The antibody was generated using a recombinant protein immunogen corresponding to human TIM-3. For applications such as WB, IF-P, IHC-P, and ELISA, consider optimizing antibody concentrations and validating specificity in the relevant sample system.
  • In WB experiments, a band of approximately 33 kDa is expected based on the predicted molecular weight. Adjust blocking and detection conditions as needed for optimal signal-to-noise ratios.
  • For cross-reactivity with mouse, verify by sequence homology and test under appropriate experimental conditions, as reactivity may vary between sample types.
