Biological Background: TIM-1/KIM-1/HAVCR Function and Localization
- TIM-1/KIM-1/HAVCR, encoded by the Havcr1 gene, is a type I transmembrane glycoprotein also known as Kidney injury molecule 1 (KIM-1) and T cell immunoglobulin and mucin domain-containing protein 1 (TIMD-1). It belongs to the TIM family of phosphatidylserine receptors.
- Acts as a phosphatidylserine receptor, playing a critical role in regulatory B-cell homeostasis, including generation, expansion, and suppressor functions.
Related references: PMID:21821911 PMID:25645598 PMID:32668241 - Functions in efferocytosis and phagocytosis: binds phosphatidylserine on apoptotic cells, facilitating their engulfment by kidney epithelial cells and also enabling phagocytosis of bacteria.
- Serves as a ligand for P-selectin/SELPLG, regulating activated T-cell trafficking during inflammatory responses, thereby controlling T-cell accumulation in the central nervous system and autoimmune disease induction.
Related references: PMID:24703780 - Regulates T-cell proliferation and expression of anti-inflammatory cytokines and co-inhibitory ligands, including IL-10.
Related references: PMID:15793576 PMID:25582854 PMID:25645598 - Subcellular localization includes the cell membrane, phagocytic cup, and cytoplasmic vesicles/phagosomes, consistent with its role in phagocytosis and signaling.
- Post-translational modifications include N-linked glycosylation and disulfide bonds, typical of immunoglobulin superfamily members, which may influence ligand binding and stability.
- Tissue-specific expression: expressed by stimulated T-cells and at higher levels on B-cells, both constitutively and after activation, with upregulation during primary antigen stimulation.
Related references: PMID:11725301 PMID:21821911
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (amino acids 22-237), which is likely recognized by the antibody; thus, it may detect both full-length and soluble ectodomain forms. Consider optimizing antigen retrieval or denaturing conditions for Western blot to detect the 33 kDa band.
- For ELISA, use indirect or sandwich ELISA with appropriate coating buffers, and validate specificity using recombinant TIM-1 protein or cell lysates from mouse T-cell or kidney epithelial cell lines.
- Cross-reactivity with human TIM-1 is reported; however, due to polyclonal nature, confirm cross-reactivity in your specific sample system and consider batch-to-batch variability.
- When studying tissue-specific expression, note that TIM-1 is upregulated upon injury or activation; therefore, include appropriate positive and negative controls.
