Biological Background: TIGAR Function and Localization

  • TIGAR (TP53-induced glycolysis and apoptosis regulator), also known as FR2BP or C12orf5, functions as a fructose-2,6-bisphosphatase that hydrolyzes fructose-2,6-bisphosphate and fructose-1,6-bisphosphate. Related references: PMID:19015259

  • It acts as a negative regulator of glycolysis by reducing intracellular fructose-2,6-bisphosphate levels in a p53/TP53-dependent manner, thereby activating the pentose phosphate pathway (PPP) and increasing NADPH production. Related references: PMID:16839880 PMID:22887998

  • By boosting NADPH levels, TIGAR promotes reduced glutathione generation, which decreases reactive oxygen species (ROS) and protects cells from oxidative stress- and metabolic stress-induced apoptosis. Related references: PMID:16839880 PMID:19713938 PMID:22887998

  • The protein localizes to the cytoplasm, nucleus, and mitochondrion, consistent with its roles in metabolic regulation and apoptosis.

  • In cancer cells, TIGAR supports survival under hypoxia by inhibiting glycolysis and shifting metabolism toward mitochondrial respiration; it also facilitates DNA repair through PPP activation during genome stress. Related references: PMID:22887998 PMID:25928429 PMID:23726973

  • TIGAR is expressed in brain, breast tumors, and glioblastomas, highlighting its relevance in both normal tissues and oncological contexts. Related references: PMID:22887998 PMID:21820150

  • Post-translational modifications include acetylation, and the protein is implicated in autophagy and apoptotic pathways.

Experimental Guidance and Technical Tips

  • For Western blot analysis, confirm the detection of a band at approximately 30 kDa, matching the predicted molecular weight of TIGAR. Verify specificity by pre-incubating the antibody with the immunizing peptide if non-specific bands appear.
  • In immunofluorescence (IF/ICC), optimize fixation and permeabilization protocols to visualize TIGAR in its expected cytoplasmic, nuclear, or mitochondrial compartments depending on cell type and stress conditions.
  • The antibody cross-reacts with mouse and rat samples; include appropriate species-specific positive and negative controls when working across these species.
  • For ELISA applications, titrate the antibody to determine optimal coating and detection concentrations, and validate using recombinant TIGAR protein or cell lysates.
