Biological Background: Nucleolysin TIAR Function and Localization
- Nucleolysin TIAR (TIAL1) is an RNA-binding protein that regulates alternative pre-mRNA splicing and the formation of cytoplasmic stress granules. Related references: PMID:10613902 PMID:1326761 PMID:17488725
- TIAR preferentially binds uridine-rich RNAs and activates splicing of exons with weak 5' splice sites followed by U-rich stretches, including autoregulation of its own pre-mRNA and that of TIA1. Related references: PMID:8576255
- TIAR acts downstream of stress-induced EIF2S1/EIF2A phosphorylation to recruit untranslated mRNAs to cytoplasmic stress granules, a key cellular stress response. Related references: PMID:10613902
- The protein possesses nucleolytic activity against cytotoxic lymphocyte target cells and may be involved in apoptosis. Related references: PMID:1326761
- TIAR is localized in the nucleus, cytoplasm, cytolytic granules, and stress granules, reflecting its dynamic role in RNA metabolism and cellular stress.
- Expression is detected in brain, heart, kidney, lung, and skeletal muscle.
- Keywords: RNA-binding, alternative splicing, apoptosis, phosphoprotein, acetylation.
Experimental Guidance and Technical Tips
- The immunogen (aa 266-375) includes part of the RNA recognition motif region, which is conserved across species, supporting the reported cross-reactivity with mouse and rat.
- For Western blot, a band around 42 kDa (observed molecular weight) is expected; consider using appropriate tissue or cell lysates expressing TIAL1.
- For IHC-P, validate with positive control tissues such as brain or heart where TIAL1 is known to be expressed.
