Biological Background: TIA1 Function and Localization
- TIA1 (T-cell-restricted intracellular antigen-1) is a cytotoxic granule-associated RNA-binding protein that regulates alternative pre-mRNA splicing and mRNA translation by binding to uridine-rich (U-rich) RNA sequences. Related references: PMID:11106748 PMID:12486009 PMID:17488725
- It modulates alternative splicing of apoptosis-related genes, such as FAS, by promoting inclusion of exon 6 to generate the pro-apoptotic membrane-bound receptor. Related references: PMID:11106748 PMID:17488725 PMID:1934064
- TIA1 is involved in translational repression by binding to AU-rich elements (AREs) in 3' UTRs of target mRNAs, including TNF and PTGS2, and participates in stress granule formation upon cellular stress.
- Alternative splicing produces at least two isoforms: a longer isoform (p40-TIA-1) and a shorter isoform, with the longer isoform displaying enhanced splicing regulatory activity.

- The immunogen (amino acids 287-386 of human TIA1) lies within a region that is highly conserved between human and mouse, supporting the observed cross-reactivity. Confirm homology for other species of interest.
- For Western blotting, the expected band is around 43 kDa, but multiple bands may appear due to alternative splicing isoforms or post-translational modifications. Validate in the relevant sample system.
- In immunofluorescence (IF/ICC) and immunohistochemistry (IHC-P), TIA1 can be detected in both nuclear and cytoplasmic pools. To visualize stress granules, consider treating cells with stressors (e.g., sodium arsenite) prior to fixation.
- Optimal dilutions and experimental conditions should be determined empirically for each application and sample type.
