Biological Background: Thromboxane synthase Function and Localization

  • Thromboxane synthase (TBXAS1), also known as cytochrome P450 5A1, is encoded by the TBXAS1 gene in humans. It is a key enzyme in the arachidonic acid cascade.
  • Catalyzes the conversion of prostaglandin H2 (PGH2) to thromboxane A2 (TXA2), a potent vasoconstrictor and inducer of platelet aggregation (PubMed:11097184, PubMed:11297515, PubMed:22735388).
    Related references: PMID:11097184 PMID:11297515 PMID:22735388
  • Also cleaves PGH2 to produce 12-hydroxyheptadecatrienoic acid (12-HHT) and malondialdehyde, a DNA damage mediator, in stoichiometric amounts equivalent to TXA2 (PubMed:11297515, PubMed:22735388, PubMed:9873013).
    Related references: PMID:11297515 PMID:22735388 PMID:9873013
  • Displays dehydratase activity toward (15S)-hydroperoxy-(5Z,8Z,11Z,13E)-eicosatetraenoate (15(S)-HPETE), converting it to 15-KETE and 15-HETE (PubMed:17459323).
    Related references: PMID:17459323
  • Localized to the endoplasmic reticulum membrane, where it functions as a monooxygenase and oxidoreductase requiring heme and iron cofactors.
  • Expressed in platelets, lung, kidney, spleen, macrophages, and lung fibroblasts, consistent with its role in hemostasis and inflammation.
  • Keywords include alternative splicing and disease variant, reflecting its association with Ghosal hematodiaphyseal dysplasia (GHOSAL) and bleeding disorder platelet-type 14 (BDPLT14).

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 355-534 of human thromboxane synthase; the antibody is expected to detect the full-length protein (~61 kDa) by Western blot.
  • For WB, test in platelet lysates or thromboxane synthase-expressing cell lines; consider using a reducing agent and appropriate blocking buffer to minimize non-specific binding.
  • For IHC-P, antigen retrieval may be necessary; validate in positive control tissues such as lung or kidney sections, and include isotype controls.
  • In ELISA, the antibody can be used as a capture or detection reagent; optimization of coating buffer and concentration is recommended.
