Biological Background: Thromboxane-A Synthase Function and Localization
- Thromboxane-A synthase (TBXAS1), also known as cytochrome P450 5A1 and hydroperoxy icosatetraenoate dehydratase, belongs to the cytochrome P450 superfamily and is primarily involved in eicosanoid metabolism.
- Catalyzes the conversion of prostaglandin H2 (PGH2) to thromboxane A2 (TXA2), a potent vasoconstrictor and platelet aggregation agonist, and co-produces 12-hydroxyheptadecatrienoic acid (12-HHT) and malondialdehyde (MDA) in stoichiometric amounts. Related references: PMID:11097184 PMID:11297515 PMID:22735388 PMID:9873013
- Exhibits dehydratase activity, converting (15S)-hydroperoxy-(5Z,8Z,11Z,13E)-eicosatetraenoate (15(S)-HPETE) to 15-KETE and 15-HETE. Related references: PMID:17459323
- Localized to the endoplasmic reticulum membrane, where it functions as a monooxygenase and isomerase, requiring heme and iron cofactors.
- Predominantly expressed in platelets, lung, kidney, spleen, macrophages, and lung fibroblasts, consistent with its roles in hemostasis and inflammation.
- Alternative splicing generates multiple isoforms; disease-associated variants are linked to Ghosal hematodiaphyseal dysplasia and bleeding disorder platelet-type 14 (BDPLT14).
- Annotated with keywords including Fatty acid biosynthesis, Lipid metabolism, Prostaglandin biosynthesis, and Reference proteome, reflecting its central role in lipid mediator production.
Experimental Guidance and Technical Tips
- The immunogen was designed as a synthetic peptide spanning the N-terminal amino acids 1–100 of human Thromboxane synthase (P24557); this region may confer target specificity across isoforms and species.
- For Western blot (WB), the protein carries a predicted molecular weight of approximately 61 kDa; consider using platelet-rich plasma, lung tissue, or macrophage lysates as positive controls.
- ELISA applications may employ direct or capture formats; validate the antibody’s performance in the relevant sample matrix and with appropriate recombinant protein standards.
- Cross-reactivity with mouse is confirmed, making this antibody suitable for translational models; optimization of dilutions and detection conditions is recommended for each application.
