- THRAP3, also known as TRAP150 and BCLAF2, is a thyroid hormone receptor-associated protein encoded by the THRAP3 gene.
- Involved in pre-mRNA splicing, where it remains associated with spliced mRNA through interactions with the exon junction complex (EJC) and can trigger mRNA decay independent of nonsense-mediated decay.
- Regulates mRNA stability, notably as a component of the SNARP complex that stabilizes cyclin D1 (CCND1) mRNA by binding its 3' end, and participates in nuclear mRNA decay.
- Functions in DNA damage response; is excluded from DNA damage sites in parallel with transcription inhibition, a process that may involve the SNARP complex.
- Although initially identified as part of the TRAP/Mediator complex, it is not a stable subunit but cooperates with HELZ2 to enhance PPARG-mediated transcriptional activation, playing a role in adipocyte differentiation.
- Acts as a coactivator of the circadian clock by promoting the transcriptional activity and target gene binding of the CLOCK-BMAL1 heterodimer. Related references: PMID:24043798
- Subcellularly localized to the nucleus, including the nucleoplasm and nuclear speckles.
