Biological Background: THOC7 Function and Localization
- THOC7 is a component of the THO subcomplex of the TREX complex, also known as fSAP24, hTREX30, and NIF3L1BP1.
- It couples mRNA transcription, processing, and nuclear export by selectively binding spliced mRNA (not unspliced pre-mRNA). Related references: PMID:15833825, PMID:15998806, PMID:17190602
- Plays a key structural role in the oligomerization of the THO-DDX39B complex. Related references: PMID:33191911
- Required for efficient export of polyadenylated RNA. Related references: PMID:23222130
- Recruited to spliced mRNAs upstream of the exon-junction complex in a splicing- and cap-dependent manner, facilitating export through the TAP/NXF1 pathway. Related references: PMID:15833825, PMID:15998806, PMID:17190602
- Localizes to the cytoplasm, nucleus, and nuclear speckles.
- Contains acetylation and phosphorylation sites, a coiled-coil domain, and is an RNA-binding protein.
- The TREX complex, including THOC7, is essential for the export of Kaposi's sarcoma-associated herpesvirus (KSHV) intronless mRNAs and infectious virus production.
Experimental Guidance and Technical Tips
- The immunogen covers the N-terminal region (aa 1-204), suggesting recognition of full-length THOC7; consider antigen retrieval or native conditions to preserve epitopes.
- For Western blot, the predicted molecular weight is ~24 kDa; validate in the relevant sample system (human, mouse, or rat) and confirm band specificity with siRNA knockdown or knockout controls.
- For ELISA, use the recombinant immunogen as a positive control and optimize coating concentrations.
- Since cross-reactivity spans human, mouse, and rat, this antibody may be suitable for comparative studies; verify species-specific reactivity in pilot experiments.
- THOC7 is part of a multi-protein complex; avoid harsh denaturing conditions that might disrupt epitope recognition; mild lysis buffers are recommended.
